During aging, various complex structural and functional changes occur in the skin, with a progressive atrophy of the dermis and epidermis. Not only is the dermis impacted, with loss and disorganization of collagen fibrils in the extracellular matrix, but so is the epidermis, with a reduced mitosis rate and a loss of cellular cohesion, and as well as the dermal-epidermal junction (DEJ) with a flattening and a less effective anchoring system.
To improve the signs of skin aging, we developed Replexium, a synergistic complex combining Acetyl Tetrapeptide-9 and Acetyl Tetrapeptide-11 with bioavailability agents, targeting extracellular matrix, epidermal, and DEJ structural integrity.
The Acetyl Tetrapeptide-9 in Replexium increased the age-affected synthesis of lumican proteoglycan and the number of dermal fibroblasts in vitro, to improve the impaired collagen fibril organization in the aging dermis. On 3D bioprinted skin, the complex of the 2 tetrapeptides improved the density and organization of the extracellular collagen fibers for a denser dermis.
Tested in vivo in a clinical test on human volunteers compared to 3% of a Peptide-blend benchmark, Replexium demonstrated a quicker effect on dermal density and similar anti-wrinkle performance. Replexium visibly improved the appearance of wrinkles and the density of the dermal structure, for a fast anti-wrinkle and skin firming efficacy within 3 weeks.
